In vitro transcribed mRNA for expression of designer nucleases: Advantages as a novel therapeutic for the management of chronic HBV infection

Abdullah Ely et al.

Summary: Chronic infection with hepatitis B virus remains a significant global medical issue, with the need for a cure becoming increasingly urgent as complications such as cirrhosis and hepatocellular carcinoma are often fatal. Current therapies are unable to eradicate the virus due to the persistence of viral cccDNA, making the development of methods to eliminate or inactivate the cccDNA a key focus of research in the quest to cure chronic hepatitis B.

Advances in lipid carriers for drug delivery to the gastrointestinal tract

Yining Xu et al.

Summary: Lipid-based formulations have reappeared as oral drug delivery systems, thanks to advances in evaluating the in vivo fate of formulations and an improved understanding of gastrointestinal processes. The use of lipid-based formulations has expanded beyond highly lipophilic drugs to include hydrophilic peptides and macromolecules. Additionally, targeted nanocarriers have been developed based on gut physiology to explore novel targets in the field of drug delivery.

Understanding the antiviral effects of RNAi-based therapy in HBeAg-positive chronic hepatitis B infection

Sarah Kadelka et al.

Summary: The RNA interference drug ARC-520 has shown significant efficacy in reducing HBV DNA, HBeAg, and HBsAg levels in patients, with a more than 96% efficacy in blocking HBsAg and HBeAg at the start of treatment, gradually decreasing to around 50% within 1-4 months. The combined use of ARC-520 and entecavir maintained a consistent efficacy of over 99.8% in reducing HBV DNA levels, with entecavir mediating continuous decline in HBV DNA and ARC-520 mediating strong but temporary reductions in HBsAg and HBeAg. The developed modeling framework could aid in assessing the development of RNAi drugs for HBV infection.

Giulia Anderluzzi et al.

Summary: Through the study of four different cationic delivery systems, it was found that they have different characteristics and effects. In vitro, particulate systems were better at promoting cell uptake and antigen processing, while emulsions were less effective. However, emulsions induced the highest antibody responses in vivo compared to other systems tested.

Invasomes: Potential vesicular systems for transdermal delivery of drug molecules

Shweta Jain et al.

Summary: Vesicular drug delivery systems, such as invasomes, have the potential to improve therapeutic efficacy by targeting drugs at a predetermined rate. While invasomes have been explored extensively for transdermal drug delivery, literature on them is relatively scarce. Key components like ethanol, terpenes, and phospholipids play a crucial role in influencing the physicochemical properties of invasomes, but their stability remains a limiting factor.
