Congenital sensorineural hearing loss as the initial presentation of PTPN11-associated Noonan syndrome with multiple lentigines or Noonan syndrome: clinical features and underlying mechanisms

Summary: Germline variants in PTPN11 can cause Noonan syndrome with multiple lentigines (NSML) or Noonan syndrome (NS), with congenital sensorineural hearing loss (SNHL) being a major symptom. Experimental studies in mice and zebrafish suggest a potential association between PTPN11 and inner ear structures, supporting the recommendation for screening PTPN11 in patients with congenital hearing loss.
