Summary: The study found that the rCBF response was reduced in early stages of 3xTgAD mice and almost absent in older mice, related to the progression of AD pathology. The inhibitor N-omega-nitro-L-arginine eliminated the rCBF response to diazoxide, indicating the involvement of NO in vasorelaxation. The PS1(M146V) mice showed reduced rCBF response to diazoxide and increased high molecular weight Abeta oligomers, suggesting PS1(M146V) contributed to the dysregulation of rCBF response in AD mice.
