When comparing miRNA and piRNA dysregulation in transgenic C. elegans nematodes overexpressing human alpha-synuclein wild-type (WT) or mutant A53T, genetic crosses with tdp-1 null mutants improved behavioral deficits and altered the number of dysregulated miRNAs and piRNAs. Prediction of target genes by specific miRNAs shed light on the molecular landscape of small non-coding RNA dysregulation in PD-associated neurodegenerative disorders.

Summary: This study revealed the mechanisms by which hypoxic cancer cells induce CAF-like differentiation through secreting sEVs and TGF-beta, with miRNAs delivered by sEVs playing a crucial role in this process. Elevated levels of miR-192/215 in HNSCC tissue-derived sEVs indicated a hypoxic and aggressive cancer stroma, and miR-215 in tumor tissue-derived sEVs was associated with poor overall survival in HNSCC patients. The findings suggest that sEVs serve as a messenger to transfer miRNAs from cancer cells to fibroblasts, contributing to the remodeling of the hypoxic tumor microenvironment and indicating the potential of cancer tissue-derived sEVs as biomarkers.

MicroRNA-124-3p suppresses PD-L1 expression and inhibits tumorigenesis of colorectal cancer cells via modulating STAT3 signaling

Summary: miR-124 is significantly downregulated in CRC tissues compared with normal samples, and its downregulation is negatively correlated with PD-L1 expression. Overexpression of miR-124 can inhibit Tregs differentiation by reducing PD-L1 expression, suppress CRC cell proliferation, induce apoptosis, and decrease migration and invasion capabilities of CRC cells. Additionally, miR-124 can target the STAT3 signaling pathway in CRC cells, suggesting its potential as a therapeutic target for colorectal cancer.

The study of extracellular vesicles (EVs), especially exosomes, in pancreatitis has shown potential for identifying cell-to-cell communication and influencing the disease's development and progression. Plasma exosomes of acute pancreatitis can reach the alveolar cavity and activate alveolar macrophages to cause acute lung injury, suggesting their use as biomarkers and therapeutic agents for this disease.
