HIV-1 Env induces pexophagy and an oxidative stress leading to uninfected CD4+T cell death

Coralie F. Daussy et al.

Summary: The immunodeficiency observed in HIV-1-infected patients is mainly due to bystander CD4(+)T lymphocyte cell death, triggered by the viral envelope glycoprotein Env-induced autophagy and oxidative stress. Env-mediated autophagy targets peroxisomes, leading to the degradation of these organelles and contributing to immunodeficiency induced by HIV-1.

Catalase deficiency induces reactive oxygen species mediated pexophagy and cell death in the liver during prolonged fasting

Raghbendra Kumar Dutta et al.

Summary: The study demonstrates that prolonged fasting in catalase-knockout mice significantly increases ROS production, leading to liver-specific pexophagy and cell death. The use of antioxidants and autophagy inhibitors can suppress the inflammatory response and disease progression.

Autophagy Inhibitors Do Not Restore Peroxisomal Functions in Cells With the Most Common Peroxisome Biogenesis Defect

Femke C. C. Klouwer et al.

Summary: This study investigated the potential use of autophagy inhibitors as a treatment option for PBD-ZSD patients carrying the PEX1-G843D mutation. However, the results showed that autophagy inhibitors did not improve peroxisomal functions, and in fact worsened metabolic functions and protein import in cells with the mutation. The study suggests that L-arginine remains a promising therapeutic compound for PBD-ZSD patients.
