Enzyme evolution for industrial biocatalytic cascades

Jovana Nazor et al.

Summary: Multi-step, biocatalytic cascades show great potential in leading the further adoption of enzymes in the chemical industry by enabling difficult chemistry under mild reaction conditions with minimal waste generation. Recent advances in biocatalysis have led to increasingly complex cascades, optimized through enzyme engineering and evolution to improve activity, selectivity, and stability. Industrially relevant enzyme cascades have been successfully developed with the aim of sustainable and economically attractive biocatalytic synthesis of complex chemicals.

Enzyme engineering for biosynthetic cascades

John A. McIntosh et al.

Summary: Biosynthetic cascades have revolutionized the approach to synthetic challenges, setting new standards for green and sustainable chemistry. While early work focused on natural products and relied on natural enzymes, the demand for synthetic derivatives and wholly synthetic molecules in pharmaceuticals and fine chemicals has led to the need for extensive protein engineering. Significant progress has been made in protein engineering for syntheses involving multiple enzyme-catalyzed steps both in vivo and in vitro from 2018 to 2020.
