One stem cell program to rule them all?

Summary: Stem cells are likely an ancestral feature of all animals, with those possessing the properties of self-renewal and multipotency potentially sharing ancestral genetic circuitry. However, identifying and comparing stemness gene signatures across diverse animals and large evolutionary distances is challenging, requiring experimental demonstration, genomic methods, and mapping gene homology. The discovery and understanding of ancestral stemness programs may ultimately shed light on the dysregulation of stemness programs in cancer stem cells.

Summary: The study revealed that glioblastomas contain diverse cell populations, with GSCs showing high transcriptional heterogeneity associated with normal neural development and inflammatory wound response. Genome-wide CRISPR-Cas9 dropout screens validated this observation. Additionally, the majority of malignant cells were found to organize along an orthogonal astrocyte maturation gradient while retaining founder GSC transcriptional programs.
