TRIM21 Is Decreased in Colitis-associated Cancer and Negatively Regulates Epithelial Carcinogenesis

Guangxi Zhou et al.

Summary: TRIM21 expression is decreased in colitis-associated cancer, and its deficiency promotes colorectal carcinogenesis, impacting tumor cell proliferation, tissue remodeling, and inflammatory cytokines. This suggests TRIM21 may be a potential therapeutic target for CAC.

Targeting TRIM Proteins: A Quest towards Drugging an Emerging Protein Class

Francesca D'Amico et al.

Summary: The ubiquitylation machinery plays a key role in various biological processes and its dysregulation is associated with multiple diseases. TRIM proteins, as a novel class of ligases for drug development, are gaining attention in the pharmaceutical field. Understanding the current research progress and potential targeting options for single polypeptide RING E3 ligases can provide insights into their therapeutic potential for diseases.

TRIM37 overexpression is associated with chemoresistance in hepatocellular carcinoma via activating the AKT signaling pathway

Guosheng Tan et al.

Summary: The study showed that TRIM37 expression was significantly increased in HCC cells and tissues, especially in Sorafenib-resistant HCC tissues. High TRIM37 expression was correlated with poor prognosis in HCC patients. TRIM37 overexpression conferred Sorafenib resistance on HCC cells, while inhibition of TRIM37 sensitized HCC cells to Sorafenib cytotoxicity. Additionally, TRIM37 upregulated AKT activity and phosphorylated AKT levels, thereby activating canonical AKT signaling.

circNDUFB2 inhibits non-small cell lung cancer progression via destabilizing IGF2BPs and activating anti-tumor immunity

Botai Li et al.

Summary: It was found that circNDUFB2 is downregulated in NSCLC tissues and inhibits the growth and metastasis of NSCLC cells by enhancing the interaction between TRIM25 and IGF2BPs. circNDUFB2 also activates anti-tumor immune responses by binding to RIG-I.

Knockdown of TRIM26 inhibits the proliferation, migration and invasion of bladder cancer cells through the Akt/GSK3β/β-catenin pathway

XiaoJuan Xie et al.

Summary: TRIM26 is upregulated in bladder cancer tissues and its absence inhibits cellular behaviors, while overexpression promotes cell proliferation, migration and invasion. TRIM26 regulates cell proliferation, migration, and invasion in bladder cancer through the Akt/GSK3 beta/beta-catenin pathway.

The Role of mTOR Signaling as a Therapeutic Target in Cancer

Nadezhda V. Popova et al.

Summary: This review summarizes the current information on the role of phosphatidylinositol-3-kinase (PI3K)/AKT/mammalian target of rapamycin (mTOR) signaling in cancer as a potential target for new therapy options. The mTOR and PI3K/AKT/mTORC1 (mTOR complex 1) signaling pathways are critical for regulating fundamental cell processes, and their deregulation is implicated in cancer, metabolic dysregulation, and aging. The review also discusses recent data on the structure and function of the mTOR pathway, mechanisms of deregulation in human cancers, and the use of PI3K/AKT/mTOR inhibitors in clinical studies for cancer treatment, including issues of resistance and adverse effects.

Obesity related microRNA-424 is regulated by TNF-α in adipocytes

Qin-Zhi Xiao et al.

Summary: Obesity is associated with inflammation in adipose tissue, where dysregulation of miR-424 has been linked to metabolic syndromes. The study found that miR-424 expression was increased in adipose tissue of obese children but decreased during adipogenesis in cultured adipocytes. TNF-alpha was shown to downregulate miR-424 expression by binding to its promoter, and bioinformatics analysis revealed target genes involved in adipogenesis pathways like Wnt signaling.
