Interactions of α-synuclein oligomers with lipid membranes

Greta Musteikyte et al.

Summary: Parkinson's disease is an incurable neurodegenerative disorder characterized by the formation of Lewy bodies. Oligomeric forms of the protein alpha-synuclein interacting with lipid membranes are considered the main pathogenic species. Understanding the structural and biophysical features of these oligomers and their interactions with membranes is crucial for deciphering the etiology of Parkinson's disease and its toxicity in vivo.

Summary: Mutations in the GBA gene, encoding GCase, are the highest genetic risk factor for Parkinson’s disease, leading to decreased GCase activity and stability which results in the accumulation of lipid substrates and αS. Changes in αS and lipid levels are correlated with alterations in GCase activity/protein levels. Current strategies aim to revert the changes in GCase activity/protein, αS, and lipid levels in the context of PD.

Folding Free-Energy Landscape of α-Synuclein (35-97) Via Replica Exchange Molecular Dynamics

Summary: The misfolding and aggregation of alpha-synuclein play a crucial role in neurodegenerative disorders, with the formation of fibrils involving various intermediates. Simulation results show that conformational rearrangements of alpha-synuclein at physiological temperature mainly exhibit alpha-helix structures. These findings provide valuable insights for understanding the formation of oligomers in neurodegenerative diseases.
