Mitochondrial DNA stress triggers autophagy-dependent ferroptotic death

This effect is mediated by zalcitabine-induced mitochondrial DNA stress and the activation of a DNA sensing pathway, leading to cell death via lipid peroxidation. Genetically or pharmacologically inhibiting this pathway reduces the anticancer effects of zalcitabine.

These findings may lead to the development of new antitumor therapy targeting autophagy-dependent cell death.
