Group 2 innate lymphoid cells in nasal polyposis

Whitney W. Stevens et al.

Summary: This study reviewed the presence, function, and activation of innate lymphoid cells (ILC2s) in chronic rhinosinusitis with nasal polyps (CRSwNP). It found that ILC2s are highly elevated and activated in nasal polyps, producing type 2 cytokines, indicating a crucial role in the pathogenesis of CRSwNP. Targeting the upstream mediators activating ILC2s and the downstream products released by these cells may improve clinical outcomes in CRSwNP.

IL-10 production by ILC2s requires Blimp-1 and cMaf, modulates cellular metabolism, and ameliorates airway hyperreactivity

Emily Howard et al.

Summary: The study identified a unique subset of ILC2s, known as ILC2(10)s, which actively produce IL-10 to suppress proinflammatory ILC2 functions through autocrine and paracrine signaling, resulting in reduced lung inflammation and airway hyperresponsiveness in vivo. Furthermore, the research highlights the metabolic dependency of ILC2(10)s on the glycolytic pathway for IL-10 production, contrasting with the conventional fatty acid oxidation pathway used for proinflammatory effector functions.

Induction of IL-10-producing type 2 innate lymphoid cells by allergen immunotherapy is associated with clinical response

Summary: IL-10(+) ILC2s play a crucial role in modulating grass-pollen allergy, attenuating Th responses, and maintaining epithelial cell integrity. Patients with grass-pollen allergy have lower levels of IL-10(+) KLRG1(+) ILC2s, but this ability to produce IL-10 is restored after immunotherapy treatment. The mechanisms underlying this involve modifications in retinol metabolic pathway, cytokine-cytokine receptor interaction, and JAK-STAT signaling pathways in the ILCs.

Summary: Different inflammatory endotypes in CRSsNP are controlled by distinct molecular mechanisms, and biomarkers in nasal lavage fluids may help distinguish tissue endotypes in CRSsNP, allowing for more precise and personalized medical treatments.
