Topological data analysis reveals genotype-phenotype relationships in primary ciliary dyskinesia

Amelia Shoemark et al.

Summary: This study used topological data analysis to investigate genotype-phenotype relationships in PCD, revealing that different gene mutations have varying effects on disease severity and presentation, and that cilia ultrastructure and beat pattern defects are closely related to specific causative gene groups.

Developing DMD therapeutics: a review of the effectiveness of small molecules, stop-codon readthrough, dystrophin gene replacement, and exon-skipping therapies

Omar Sheikh et al.

Summary: Currently, steroids are the most commonly used medication for DMD patients, while stop codon readthrough, gene replacement, and exon skipping therapies aim to restore dystrophin expression. Among these strategies, gene replacement therapy has gained recent momentum, and exon-skipping continues to be popular. FDA approval of three exon-skipping antisense oligonucleotides demonstrates regulatory progress, but controversy remains regarding the effectiveness and design of eteplirsen. Cell-penetrating peptides show promise in treating DMD-related cardiomyopathy but pose regulatory challenges.

Motile cilia and airway disease

Marie Legendre et al.

Summary: The article discusses the role of regulated airway epithelial development in the differentiation of motile ciliated cells in the human respiratory tract, as well as ciliary dysfunction caused by genetic mutations. Advances in understanding respiratory epithelial development and ciliary biology have the potential to improve disease diagnosis, management, and family counseling.

Defects in the cytoplasmic assembly of axonemal dynein arms cause morphological abnormalities and dysmotility in sperm cells leading to male infertility

Isabella Aprea et al.

Summary: Male fertility issues worldwide may be caused by defects in the preassembly of outer and inner dynein arms, leading to abnormal sperm flagella, impaired sperm motility, and infertility. Studying sperm cells of individuals with mutations in genes related to this process helps understand the clinical relevance of preassembly of dynein arms on male fertility.
