The roles ofPAD2-andPAD4-mediated protein citrullination catalysis in cancers

Yanbin Wang et al.

Summary: Peptidylarginine deiminases (PADs) are enzymes that play important roles in biological processes by converting arginine to citrulline. They are crucial in physiological and pathological processes related to cancer, with PAD2 and PAD4 exhibiting specificity in various types of cancer. Studying PADs and their inhibitors may aid in disease diagnosis and treatment.

PADs in cancer: Current and future

Yu Zhang et al.

Summary: PADs are calcium-dependent enzymes that catalyze the conversion of arginine residues into citrulline, leading to the formation of citrullinated proteins with altered structure and function associated with various pathological diseases, including inflammation and autoimmune diseases. Studies have shown that PADs are implicated in cancer progression, but their functions in tumors are complex due to involvement in various signaling pathways.

Peptidylarginine Deiminase Inhibition Prevents Diabetes Development in NOD Mice

Fernanda M. C. Sodre et al.

Summary: In this study, systemic inhibition of peptidylarginine deiminases (PADs) through BB-Cl-amidine treatment prevented diabetes development in NOD mice by reducing citrullination levels in the pancreas, decreasing circulating autoantibody titers, and reducing neutrophil extracellular trap formation. Additionally, the treatment induced a shift in cytokine production and increased regulatory T cell frequency. This highlights the potential of PAD inhibition for treating autoimmune diseases like type 1 diabetes.

Extracellular DNA Correlates with Intestinal Inflammation in Chemically Induced Colitis in Mice

Martin Maronek et al.

Summary: Circulating extracellular DNA (ecDNA) exacerbates many diseases, with higher concentrations found in inflammatory conditions like inflammatory bowel disease (IBD). A study using a mice model of chemically induced colitis found that plasma ecDNA concentration peaked on the seventh day of dextran sulfate sodium (DSS) intake. Early administration of PAD4 inhibitors did not alleviate disease activity but did reduce ecDNA concentration.
