Sequential Prostate Magnetic Resonance Imaging in Newly Diagnosed High-risk Prostate Cancer Treated with Neoadjuvant Enzalutamide is Predictive of Therapeutic Response

Fatima Karzai et al.

Summary: For patients with localized intermediate- or high-risk prostate cancer, baseline mpMRI can help predict pathologic response, with low relative tumor burden at initial scan being most predictive of minimal residual disease or pathologic complete response. Tailoring patient selection based on these metrics may lead to improved outcomes in high-risk disease.

Genomic and phenotypic heterogeneity in prostate cancer

Michael C. Haffner et al.

Summary: Prostate cancer is heterogeneous, with primary tumors being multifocal and metastatic tumors potentially originating from a single clone but exhibiting subclonal heterogeneity at the genomic, epigenetic, and phenotypic levels. The complex heterogeneous constellation of molecular alterations poses challenges for diagnosis and treatment of prostate cancer.

Outcomes of Post-Neoadjuvant Intense Hormone Therapy and Surgery for High Risk Localized Prostate Cancer: Results of a Pooled Analysis of Contemporary Clinical Trials

Rana R. McKay et al.

Summary: This study reports on the post-radical prostatectomy outcomes of patients who underwent intense neoadjuvant androgen deprivation therapy prior to the surgery. It found that exceptional pathological response following neoadjuvant therapy had a favorable impact on biochemical recurrence, while PTEN loss and intraductal carcinoma were associated with biochemical recurrence. Additional follow-up is needed to evaluate long-term outcomes.

The Role of Magnetic Resonance Imaging and Positron Emission Tomography/Computed Tomography in the Primary Staging of Newly Diagnosed Prostate Cancer: A Systematic Review of the Literature

Raisa S. Abrams-Pompe et al.

Summary: The study found that for newly diagnosed PCa, the sensitivity of imaging modalities such as MRI varied widely for T staging, while PSMA PET/CT showed promising results for N and M staging. However, due to data heterogeneity, small sample size, and low event rates, there is still a high level of uncertainty surrounding these findings.
