Trout and Human Plasma Protein Binding of Selected Pharmaceuticals Informs the Fish Plasma Model

Summary: Concerns are rising over the potential risks that pharmaceuticals discharged into the environment may pose to non-target organisms, such as fish. A study has found that the fish plasma model, which uses human therapeutic blood plasma concentrations to estimate the effects in fish, fails to consider the differences in plasma protein binding between fish and humans. By measuring the binding of 12 active pharmaceutical ingredients to rainbow trout and human plasma, the study reveals that for acidic APIs, the affinity to trout plasma is much lower than to human plasma, potentially leading to higher bioavailability in trout. This suggests that read-across approaches like the fish plasma model should consider differences in plasma protein binding to avoid under- or overestimation of effects in fish.
