Structural interaction between DISC1 and ATF4 underlying transcriptional and synaptic dysregulation in an iPSC model of mental disorders

Summary: Psychiatric disorders are complex mental conditions influenced by genetic and environmental factors. This study reveals that the ATF4 transcription factor, a binding partner of DISC1, is more abundant in the nucleus of DISC1 mutant neurons and plays a role in dysregulated gene expression. The research demonstrates that the mutation of DISC1 disrupts the normal interaction between DISC1 and ATF4, resulting in excessive ATF4 binding to DNA targets and deregulated gene expression, shedding light on the molecular and structural basis of transcriptional and synaptic dysregulation in mental disorders.

Pharmacological rescue in patient iPSC and mouse models with a rare DISC1 mutation

Summary: This study identified elevated PDE4 signaling in human cortical neurons differentiated from iPSCs with a DISC1 mutation, and demonstrated that inhibition of PDE4 or activation of the cAMP signaling pathway can rescue synaptic deficits. A knock-in mouse model carrying the same patient mutation in the Disc1 gene showed similar results, indicating a crucial role of the PDE4 signaling pathway in synaptic, social, and cognitive behavioral deficits.
