Novel Kras-mutant murine models of non-small cell lung cancer possessing co-occurring oncogenic mutations and increased tumor mutational burden

Summary: Conditional genetically engineered mouse models of non-small cell lung cancer (NSCLC) with increased tumor mutational burden (TMB) were established through in vitro exposure to an alkylating agent, leading to enhanced anti-tumor T cell responses and improved anti-PD-1 efficacy. However, a distinct immunosuppressed tumor microenvironment in certain cell lines limited the efficacy of PD-1 blockade, mirroring findings in human NSCLC with similar genetic characteristics. These novel models may aid in the development of innovative immunotherapies in NSCLC.
