The Role of VPS35 in the Pathobiology of Parkinson's Disease

Jenny Sassone et al.

Summary: Mutations in the VPS35 gene have been linked to late-onset familial Parkinson's disease, potentially affecting lysosomal and autophagy function, mitochondrial homeostasis, neurotransmission, and interacting with other genes associated with PD such as alpha-SYNUCLEIN, PARKIN, and LRRK2.

HES1 promotes breast cancer stem cells by elevating Slug in triple-negative breast cancer

Xiaoying Li et al.

Summary: The study indicates that HES1 promotes the stemness properties of BCSC in TNBC by upregulating Slug. TNBC patients with high levels of HES1 and Slug have worse prognosis. HES1 acts as a novel transcriptional activator for Slug, providing new insights for identifying promising prognostic biomarkers and therapeutic targets in TNBC.

Mitochondrial and Clearance Impairment in p.D620N VPS35 Patient-Derived Neurons

Zoe Hanss et al.

Summary: This study investigated the impact of the p.D620N VPS35 mutation on autophagy-lysosome pathway and mitochondrial function in induced pluripotent stem cell-derived neurons from a patient. Results showed decreased autophagic flux and lysosomal mass, accumulation of alpha-synuclein, and mitochondrial dysfunction in patient-derived neurons. This provides novel insights for potential pharmacological interventions targeting these pathways in Parkinson's disease.
