Summary: Efficient generation of highly active Streptococcus pyogenes Cas9 (SpCas9) ribonucleoprotein virus-like particles (VLPs) was achieved through specific interactions, resulting in decreased off-target insertions and deletions. VLPs containing Cas9 from different species and targeting multiple genes were found to be more efficient than combinations of single-target VLPs for simultaneous genome editing of multiple genes.
