The LARP1 La-Module recognizes both ends of TOP mRNAs

The La-related proteins: structures and interactions of a versatile superfamily of RNA-binding proteins

Anne-Catherine Dock-Bregeon et al.

Summary: The La-related proteins (LaRPs) are an ancient superfamily of RNA-binding proteins that orchestrate the main fates of RNA through a complex network of protein-RNA and protein-protein interactions. Recent studies have revealed that LaRPs have evolved unique mechanisms of RNA recognition involving structured domains and disordered regions, working together to achieve RNA substrate discrimination. This review highlights our current understanding of the structure-function relationship within the LaRP superfamily.

LARP1 isoform expression in human cancer cell lines

Hagen Schwenzer et al.

Summary: LARP1 is an oncogenic RNA-binding protein essential for ribosome biogenesis and cancer cell survival. LI-LARP1 is found to be the dominantly expressed form, while SI-LARP1 is epigenetically repressed and this repression is evolutionarily conserved in most mammalian species except a small subclade. The cell's ability to modulate splicing and expression of multiple LARP1 mRNA isoforms suggests contextually specific mechanisms of LARP1 expression.

Stabilize and connect: the role of LARP7 in nuclear non-coding RNA metabolism

Daniele Hasler et al.

Summary: La and La-related proteins (LARPs) have a common RNA interaction platform, the La module, allowing them to play crucial roles in various aspects of RNA biology. LARP7 protein binds to 7SK RNA to form 7SK snRNP, inhibiting the transcriptional activity of Pol II and also influencing the assembly of other RNPs, as well as modification, processing, and cellular transport of RNA. Reduced levels of functional LARP7 are associated with cancer and Alazami syndrome, suggesting the importance of LARP7 in regulating cellular processes and disease development.

Controversies around the function of LARP1

Andrea J. Berman et al.

Summary: LARP1 has attracted attention for its involvement in the mTOR signaling cascade and regulation of TOP mRNA translation. However, there are conflicting findings on whether LARP1 represses or activates TOP translation. In this study, opinions from four leading scientists in the field are presented to discuss these contradictory findings.

mTORC1 promotes TOP mRNA translation through site-specific phosphorylation of LARP1

Jian-Jun Jia et al.

Summary: The study reveals that mTORC1 controls TOP mRNA translation repression activity through phosphorylating LARP1, with some residues being especially sensitive to rapamycin. This phosphorylation affects the RNA-binding and translation inhibitory activities of LARP1, providing insights into a new model of translation control involving differentially regulated regions of LARP1.

The isolated La-module of LARP1 mediates 3′ poly(A) protection and mRNA stabilization, dependent on its intrinsic PAM2 binding to PABPC1

Sandy Mattijssen et al.

Summary: The La-module, consisting of a La motif (LaM), a linker, and an RNA recognition motif (RRM), is present in several La-related proteins with distinct activities in RNA metabolism. La protein and LARP7's La-modules protect nuclear RNAs with UUU-3MODIFIER LETTER PRIME tails, while LARP4 stabilizes mRNA by binding poly(A) and PABPC1. LARP1, with a PAM2 motif, exhibits similar activities mediated by its La-module, providing poly(A) protection and stabilization to mRNAs.
