Identification and elimination of genomic regions irrelevant for magnetosome biosynthesis by large-scale deletion in Magnetospirillum gryphiswaldense

Summary: This study demonstrates the use of allelic replacement method based on homologous recombination for large-scale genome editing in Magnetospirillum gryphiswaldense. Through analysis of 24 deletion mutants, genes and regions inside and outside the magnetosome island irrelevant for magnetosome biosynthesis were identified. The findings suggest that a contiguous stretch of about 100 kb could be functionally substituted by a compact 38 kb cassette, which will be beneficial for future large-scale genome editing and genetic engineering of magnetosome biosynthesis.
