Withanone and Withaferin-A are predicted to interact with transmembrane protease serine 2 (TMPRSS2) and block entry of SARS-CoV-2 into cells

Vipul Kumar et al.

Summary: A study examined the binding potential of natural compounds Withaferin-A, Withanone, and caffeic acid phenethyl ester to the cell surface receptor TMPRSS2 using molecular docking and molecular dynamics simulations. The results showed that Withanone had stronger interactions with TMPRSS2 and was able to induce changes in its allosteric site. Furthermore, Withanone downregulated TMPRSS2 mRNA expression, suggesting its potential dual action in blocking SARS-CoV-2 entry into host cells.

Structural and simulation analysis of hotspot residues interactions of SARS-CoV 2 with human ACE2 receptor

Ganesh Kumar Veeramachaneni et al.

Summary: The study reveals structural changes and interactions between nCoV S-protein RBD and ACE2 receptor, with mutations leading to increased stability and rigidity in the binding process. Key amino acid residues identified as hotspots in the interaction include Glu 35, Tyr 83, Asp 38, Lys 31, Glu 37, His 34 in ACE2 receptor and Gln 493, Gln 498, Asn 487, Tyr 505, and Lys 417 in nCoV S-protein RBD. These findings may aid in the development of blockers targeting the nCoV spike protein RBD.

Abdul Basit et al.

Summary: The study introduced a truncated version of human ACE2, tACE2, as a potential therapeutic target for Covid-19. Analysis showed that tACE2 has higher binding affinity for the receptor binding domain of SARS-CoV-2 and forms a more stable complex with it compared to intact ACE2, making it a promising candidate for drug design against Covid-19.
