Pharmacoinformatics and molecular dynamics simulation studies reveal potential covalent and FDA-approved inhibitors of SARS-CoV-2 main protease 3CLpro

Mubarak A. Alamri et al.

Summary: The study identifies potential covalent inhibitors through a structure-based approach and repurposes FDA-approved drugs against SARS-CoV-2 3CLpro. Molecular dynamics simulations characterize the binding mechanism and dynamic stability between the compounds and the protease. Identified compounds show promise as potential inhibitors for further development as COVID-19 drugs, including FDA-approved anti-hepatitis-C virus drugs paritaprevir and simeprevir.

In silicovirtual screening, characterization, docking and molecular dynamics studies of crucial SARS-CoV-2 proteins

Summary: This study aims to find effective natural compounds to inhibit the activity of the SARS-CoV-2 virus causing COVID-19. Through virtual screening and molecular docking, Baicalin and Limonin were identified as potential lead molecules, with Baicalin targeting NSP4, NSP15, and RdRp proteins, and Limonin inhibiting the human ACE2 receptor. The combination of Baicalin and Limonin is proposed for further in vitro and in vivo studies against COVID-19.

Computational analysis of dynamic allostery and control in the SARS-CoV-2 main protease

Igors Dubanevics et al.

Summary: The COVID-19 pandemic caused by the novel coronavirus SARS-CoV-2 has no publicly available vaccine or antiviral drugs at the time of writing. Research has focused on finding inhibitors for the main protease, with this study using an elastic network model to analyze the dynamics of the SARS-CoV-2 M-pro protein and suggesting potential new regions for non-competitive inhibition.
