Mitochondrial Potentiation Ameliorates Age-Related Heterogeneity in Hematopoietic Stem Cell Function

Els Mansell et al.

Summary: This study demonstrates that mitochondrial membrane potential (MMP) plays a crucial role in old hematopoietic stem cells (HSCs) and its pharmacological manipulation can alter transcriptional programs with beneficial consequences for function. Enhancing MMP in old HSCs can increase engraftment potential upon transplantation and reverse myeloid-biased peripheral blood output.

Limited rejuvenation of aged hematopoietic stem cells in young bone marrow niche

Wakako Kuribayashi et al.

Summary: As hematopoietic stem cells (HSCs) age, they exhibit reduced regenerative capacity and myeloid-biased differentiation. The aging of the HSC niche also contributes to these functional alterations. However, transplanting aged HSCs into young mice revealed that the young niche was insufficient to fully rejuvenate HSC functions, emphasizing the importance of age-associated cell-intrinsic defects in HSC aging.

Ageing and mesenchymal stem cells derived exosomes: Molecular insight and challenges

Mahdi Ahmadi et al.

Summary: Ageing is a significant risk factor for various degenerative diseases, affecting the senescence and function of Mesenchymal Stem Cells (MSCs) and their exosomes. Exosomes derived from aged MSCs exhibit accelerated biogenesis, altered cargoes, and reduced regenerative potential, potentially impacting recipient cell function negatively. Further research is required to elucidate the detailed mechanisms and translation of preclinical results into clinically relevant exosome therapies for combating ageing.
