Inducible de novo expression of neoantigens in tumor cells and mice

Summary: A new method called NINJA has been developed to control the expression of neoantigens, allowing for studies of immune responses, particularly in anti-tumor immunity. This approach bypasses central and peripheral tolerance mechanisms, inducing strong T cell responses in peripheral tissues.

Ultraviolet radiation drives mutations in a subset of mucosal melanomas

Summary: By analyzing 10 mucosal melanomas of conjunctival origin, the study found that mucosal melanomas with a predominance of UVR-associated single base substitution signature 7 (SBS7) have genomic patterns similar to cutaneous melanomas, suggesting they should not be excluded from treatments currently used for common cutaneous melanoma. While mucosal melanomas are not typically thought to be driven by UV radiation, some do present mutations characteristic of UVR exposure, indicating a potential overlap between certain mucosal and cutaneous melanomas in terms of genetic characteristics.

Summary: The study established a humanized mouse melanoma model and found high expression of chemokines in specific tumor regions, which is associated with resistance to PD-1 therapy. Further investigations showed reduced HLA-class I expression and CD8/Granz B+ T cell homeostasis in tumor areas where FOXP3 regulatory T cells and mast cells co-localize.
