Advances in Activity-Based Sensing Probes for Isoform-Selective Imaging of Enzymatic Activity

Summary: Activity-based sensing (ABS) has emerged as a powerful approach for monitoring small-molecule and enzyme activities within living systems, with a recent focus on increasing selectivity to monitor a single enzyme or isoform. The highest degree of selectivity is required for differentiating between isoforms, where the targets display significant structural similarities as a result of gene duplication or alternative splicing. This Minireview highlights key examples of small-molecule isoform-selective probes, focusing on the relevance of isoform differentiation, design strategies to achieve selectivity, and applications in basic biology or in the clinic.

Monitoring the Dynamics of Proteome Aggregation in Live Cells Using a Solubilized and Noncovalent Analogue of Fluorescent Protein Chromophores

Summary: The noncovalent, cell-permeable, and fluorogenic sensor reported here can reversibly bind to proteome amorphous aggregates in live cells, allowing for dynamic detection of both the formation and clearance of aggregated proteome. This sensor is optimized from fluorescent protein chromophores, enabling it to detect the transition from liquid-to-liquid phase separation to liquid-to-solid phase separation in cellular stresses. Overall, this sensor serves as a facile tool to screen therapeutic drugs and identify cellular pathways that ameliorate pathogenic proteome aggregation in live-cell models.
