Smoking Alters Inflammation and Skeletal Stem and Progenitor Cell Activity During Fracture Healing in Different Murine Strains

Summary: Smoking increases the risk of delayed or nonunion after fracture repair, with molecular mechanisms poorly understood. Murine models mimicking fracture healing in smokers are essential to study local cellular and molecular changes during fracture healing. Smoking affects the fracture healing capacity, chondrogenesis, skeletal stem cell activity, and inflammatory response.

Trb3 controls mesenchymal stem cell lineage fate and enhances bone regeneration by scaffold-mediated local gene delivery

Summary: The study found that Trb3 promotes osteoblastic commitment of MSCs while inhibiting adipocyte differentiation, regulating cell-fate choice through BMP/Smad and Wnt/beta-catenin signals. Local delivery of Trb3 using a GelCA-PLGA scaffold stimulated bone regeneration and inhibited fat-filled cyst formation, demonstrating a potential therapeutic strategy for improved skeletal regeneration and treatment of bone-loss disease.

Various engineering strategies are being developed to recruit, activate, and modulate the phenotype transition of macrophages to enhance the performance of engineered bone tissue.
