A perspective of our work on the development of practical metal-mediated carbon-carbon and carbon-heteroatom bond forming processes within a modern drug portfolio is presented. The case studies selected from our laboratories highlight the synthetic challenges and opportunities presented by pharmaceutically relevant scaffolds. In many cases, the target structures challenged available synthetic methods and inspired the invention of enabling technology to accelerate drug development. Additionally, considerations for running metal-mediated processes on large scale are discussed.
