In the present investigation, we have designed synthesis of a set of potential cytotoxic agents contains pyrazoyl naphthoquinone framework, from 2-hydroxy-1,4-naphthaquionone, ethylacetoacetate, substituted phenylhydrazines and aromatic aldehydes in presence of different catalysts in one-pot four component method. Various metal triflate, lewis acids and metal oxides were tested in multi-component reaction, among them, V2O5 acts as best catalyst. We have also optimized the synthetic conditions such as catalyst loading, solvents and reaction time to achieve maximum yield. It is noteworthy to mention here that the sustainable solvents like ethanol found to be suitable solvent. Further, the synthesized compounds have been employed for their in-vitro anticancer activity against cervical cancer cell line (HeLa). Among the other compound, 5i has displayed highest activity in comparison with control drug (doxorubicin) with corresponding IC50 equal to 2.9 mu M.
