Background: Rational design methods can be used to create chimeric enzymes with novel catalytic combinations. Results: Bifunctional enzymes combining xylanase and laccase activities showed enhanced catalytic activity and stability. Conclusion: Formation of an inter-domain interface alters enzyme conformation that enhances catalytic performance of the chimera. Significance: Deeper understanding of structural principles of protein fusion can improve the design of novel catalysts.
