This work describes a device for detecting E. coli bacteria by manipulating superparamagnetic beads to a sensing area and immobilizing them in a trapping well. The trapping well replaces the biochemical immobilization layer, which is commonly used in magnetic biosensor systems. A concept exploiting the volume difference between bare magnetic beads and magnetic bead-bioanalyte compounds is utilized to detect E. coli bacteria. Trapped beads are detected by the help of a tunnel magneto-resistive sensor. Frequency modulation is employed, in order to increase the signal-to-noise ratio, enabling the detection of individual superparamagnetic beads of 2.8 mu m in diameter. Replacing the biochemical immobilization layer by the trapping well greatly simplifies the detection process. After applying the mixture of E. coli and magnetic beads to the biosensor system, bacteria detection is achieved in a single step, within a few minutes.
