Description:
Abstract RNAi can wipe out the disease-causing proteins from being translated. Now RNAi technique has become a powerful tool for basic research to selectively knock down gene expression in vitro and in vivo. At the same time, both scientific and industrial communities started to develop RNAi therapeutics as the next class of drugs for treating a variety of genetic disorders, such as cancer and other diseases that are particularly hard to be addressed by current treatment strategies. Small interfering RNA (siRNA), a 21–23 nt double-stranded RNA responsible for post-transcriptional gene silencing, has attracted great interests as promising genomic drugs, due to its strong ability to silence target genes in a sequence-specific manner. In order to develop siRNAs as therapeutic agents for cancer treatment, delivery strategies for siRNA must be carefully designed and potential gene targets carefully selected for optimal anti-cancer effects. In this review, various modifications and delivery strategies for siRNA delivery are discussed. In addition, we present current thinking on target gene selection in major tumor types.

A Review: Metal Nanoparticles and their Safety Processing in Functional Foods
Metal Nanoparticles Safety Processing Functional Foods
Authors: Loutfy H. Madkour
Journal: Journal of Chemical Science and Chemical Engineering 1(3): 18-36.
Description:
Abstract Safety of nanotechnology in food industries has been discussed. Different nonmaterial’s (NMs) such as nanopowders, nanotubes, nano-fibers, quantum dots, and metal and metal-oxide nanoparticles are globally produced in large quantities due to their broad applicability in food-related industries. We present some uses of nanoparticles in food and related industries and their possible side effects. The various aspects of nanoparticles and their impact on human exposure, safety, and environmental concerns have been discussed. ZnO NPs are listed as a kind of safe substance by the FDA. The safety of nanostructured synthetic amorphous silica (SAS) as a food additive (E 551) has been discussed. and has always been produced by the same two production methods. Bioavailability increases for iron of nanometer size. The characteristics of broad spectrum of activity against food pathogens of silver nanostructures gives an insight for their potential applicability in incorporation of food packaging materials and antimicrobials for stored fruits and foods. Pharmacokinetics, Bio Distribution and Therapeutic Applications of Recently-Developed siRNA and DNA Repair Genes Recurrence
RNA interference Small interfering RNA Cancer Delivery Pharmacokinetics;

Journal: Austin J Anal Pharm Chem. 2021; 8(2): 1135.
Description:
Up regulation of cell cycle-regulating and DNA repair genes appears to have a negative impact on recurrence-free survival in patients with papillary thyroid cancer. Furthermore, recurrence is associated with thyroid dedifferentiation. Most cases address local applications or diseases in the filtering organs, reflecting remaining challenges in systemic delivery of siRNA. Small Interfering RNA (siRNA) is a promising drug candidate, expected to have broad therapeutic potentials toward various diseases including viral infections and cancer. With recent advances in bio conjugate chemistry and carrier technology, several siRNA-based drugs have advanced to clinical trials. The difficulty in siRNA delivery is in large part due to poor circulation stability and unfavorable pharmacokinetics and bio distribution profiles of siRNA. In this research we describe the pharmacokinetics and bio distribution of siRNA Nano medicines, focusing on those reported in the past 5 years, and their pharmacological effects in selected disease models such as hepatocellular carcinoma, liver infections, and respiratory diseases. The examples discussed here will provide an insight into the current status of the art and unmet needs in siRNA delivery.

Description:
Springer International Publishing, Jan 12, 2020 - Technology & Engineering - 696 pages This book provides insights and tools for better understanding redox biology and medicine and, in the long run, to finding new therapeutic strategies to target dysregulated redox processes in various diseases. It presents the recent advances in new nanomedication technologies of the effects of nanoparticles NPs on oxidative stress, RONS and ER stress. The book comprises 13 chapters covering ecotoxicology, cytotoxicity, nanotoxicity and genotoxicity mechanisms causing by the role and interactions of nanoparticles and free radicals with (RONS) and (ER) stress. Endoplasmic Reticulum (ER) Stress as a mechanism for NPs induced toxicity has been discussed. The advances of nanotechnology and the effects of nanoparticles on oxidative stress, ROS and ER stress parameters are discussed. Antioxidants, therapeutic options and regulation of the immune responses are explained throughout the book.
